  • Amisulpride, Olanzapine, Paliperidone, Quetiapine, Risperidone, Sertindole, Sulpiride, Ziprasidone, Zotepine, Clozapine.

  • Significantly lower propensity to produce EPSE and cause sustained hyperpolactinemia at clinically effective doses

  • Various modes of action, cf. typicals ? D2/5HT (no longer supported).

  • Less EPSEs  than first generation, modestly increased efficacy with olazapine, marked increased efficacy with clozapine.

  • Previously thought to be superior for negative symptoms, however research has shown that this is not the case.

  • Generally - less EPSE but higher incidence of metabolic side effects than ‘typicals’ but exceptions- aripiprazole.
